Endotoxin contamination of some erythropoietin preparations: an emerging question.

چکیده

We appreciate Dr. Drewinko’s clarification of the term “aspirate” as used in our report. Although the technique of the aspirate smear preparations was described in Materials and Methods, the more precise term “aspirate smear preparation” would be more appropriate. The experience by others as described by Dr. Drewinko has in general demonstrated the superiority of cellularity determination using the marrow aspirate clot section technique as opposed to the smear or spread preparation. We have not routinely employed the former technique, however, preferring the routine use of the biopsy for nearly all marrow examinations at our institution. Furthermore, our neport concerned the results of marrow cellularity

Contamination of erythropoietin by endotoxin: in vivo and in vitro effects on murine erythropoiesis.

Endotoxin was detected in all erythropoietin preparations tested and was removed from four lots, without loss of erythropoietic activity, by adsorption with limulus amebocyte lysate. Comparison of adsorbed (endotoxin-depleted) and nonadsorbed (endotoxin-containing) erythropoietin preparations demonstrated significant inhibition of CFU-e and BFU-e in vitro by nonadsorbed erythropoietin at concen...

The biological properties of endotoxin-free human erythropoietin.

The biological effects formerly attributed to erythropoietin were generally observed using impure preparations of the hormone containing, among the impurities, bacterial endotoxin, a contaminant that has recently been shown to affect the haemopoietic system. Erythropoietin, purified to apparent homogeneity and freed of trace contamination by endotoxin (1) promotes erythroid burst formation from...

Endotoxin Contamination of Large Volume Parenterals as Detected by Limulus Amebocyte Lysate as an Alternative to Rabbit Pyrogen Test

The possibility of replacing the rabbit pyrogen test by the Limulus Amebocyte lysate (LAL) test, as a final release test for Large Volume Parenterals (LVPs) was investigated. The sensitivity and specificity of the LAL test and rabbit pyrogen assay were studied by means of artificially contaminated parenterals. Various LVPs (mostly containing electrolytes) were spiked with 0.5 and 5 EUlml o...
